Reevaluation of Reserpine, Guanethidine, Desmethylinipramine and Cocaine as Potentiating Agents of Pressor Action of Norepinephrine

Abstract
1. The pressor responses of whole rabbits to norepinephrine, epinephrine, and angiotensin were investigated after infusions of minute doses of reserpine, guanethidine, desmethylimipramine and cocaine (reserpine: 0.05 ?g/kg/min, the other agents: 0.1 ?g/kg/min) for 2 to 3 hours. These agents potentiated the pressor responses to norepinephrine and epinephrine but not to angiotensin.
2. The pressor responses of spinal rabbits to norepinephrine and angiotensin were investigated after infusions of minute doses of reserpine (0.05?g/kg/min), guanethidine (0.2 ?g/kg/min) and desmethylimipramine(0.1 ?g/kg/min) nor 2 to 3 hours. Guanethidine and desmethylimipramine potentiated the pres or responses to norepinephrine but not to. angiotensin. Reserpine did not affect the responses to both agents.
3. The potentiated responses to norepinephrine of whole rabbits by infusions lOf the- minute doses, of reserpine, guanethidine and desmethylimipramine were further increased after intravenous injections of reserpine (0, 5 mg/kg), guanet4i-dine (10 mg/ks) and desmethylimipramine (3 mg/ky) respectively.
4. The potentiated responses to norepinephrine of spinal rabbits by infusions of the minute doses of guanethidine and desmethylimipramine were further augmented after intravenous injections of guanethidine (10 mg/kg) and desm6thylimipramine (3 mg/kg) respectively. .
5. It is inferred that the potentiation of the norepinephrine effect by infusions of the minute doses of reserpine , guanethidine, desmethylimipramine and, cocaine is produced by affecting the adrenergic receptors in the vascular smooth muscle.
